What does “DNA-free” mean?
“DNA-free” products by Molzym are defined as reagents and plastics being
negative for bacterial and fungal DNA for up to 40 PCR cycles using broad-range
bacterial and fungal PCR assays (Mastermix 16S Complete and Mastermix 18S
Complete).
Therefore, a negative control consisting of DNA-free water is used
as a template for all PCR assays/reagents.
For the MolYsis™-based kits, SU
buffer is used instead of a sample to test the whole process starting with DNA
isolation, followed by amplification and detection.

Which reagents can bear contamination by microbial DNA?
In principle, all! In particular, in our experience, the Taq DNA polymerase and primers are mostly prone to DNA contamination due to the manufacturing processes. Molzym has invested great efforts to develop a purification process for the elimination ...
What kind of consumables are to be used?
DNA-free pipette tips with filters are obligated to avoid carryover contamination during DNA preparation. Also, DNA-free 1.5 and 2.0 ml polypropylene tubes, as long as not supplied with MolYsis™ kits, should be used. Many manufacturers of plastic ...

How many PCR reactions are needed per patient?
PCR Setup / PCR reactions per sample Every sample is applied to 3 (three) master mixes: MA Bac for 16S, MA Yeasts for 18S, and MA Control for the extraction control. All master mixes need at least one negative control and MA Bac and MA Yeasts have ...
How to interpret the PCR results?
PCR results Interpretation Amplicons can either be analyzed by conventional gel electrophoresis with bands displaying the size of the amplicons (460bp for the MA Bac and 310bp for the MA Yeasts). Alternatively, a real-time PCR cycler can be used, and ...
