How many sequencing reactions are needed per sample?
Sequencing
- For
every sample with a positive 16S PCR, there are two sequencing reactions based
on two sequencing primers, SeqGN16 and SeqGP16. This allows the
identification of up to two pathogens, mainly separated into Gram-negative and
Gram-positive pathogens. But both sequencing primers
show exceptions, a few Gram-negative species will be detected by the SeqGP16, and some Gram-positive species
will be detected by the SeqGN16. The
exceptions of the sequencing primers are listed here.
- For
every sample with a positive 18S PCR, SeqYeast18 is used for sequencing.

PCR Setup / PCR reactions per sample Every sample is applied to 3 (three) master mixes: MA Bac for 16S, MA Yeasts for 18S, and MA Control for the extraction control. All master mixes need at least one negative control and MA Bac and MA Yeasts have ...
Do the primers differentiate between Gram-positive and Gram-negative bacteria?
The Set of Eubacterial Sequencing primers contains one primer, SeqGN16, which mainly targets Gram-negative bacteria, and a second one, SeqGN16, which mainly targets Gram-positive bacteria. But both sequencing primers show exceptions, a few ...
How can bacteria and fungi be identified when detected by 16S or 18S rDNA PCR?
Identification from positive amplicons The direct way is (Sanger) sequence analysis. Amplicons are purified in a simple procedure, using a commercial PCR purification kit (e.g. QIAQuick PCR Purification Kit, Qiagen, Germany). The purified amplicon is ...
Why is broad-range 16S rDNA PCR a good option for the detection of bacteria?
Broad-range 16S rDNA PCR is a favorable way of detecting bacteria in samples due to two reasons. First, the use of highly conserved regions in the 16S rRNA gene allows the amplification of sequences from any bacterial strain. Mastermix 16S Complete ...
For which application do I need the Set of Eubacterial Sequencing Primers?
Set of Eubacterial Sequencing Primers SeqGP16 & SeqGN16 The two primers are used in conjunction with Mastermix 16S Complete. Both primers bind to conserved sites within the 16S rRNA gene region amplified in the assays. Primer SeqGP16 is ...
