How many PCR reactions are needed per patient?
PCR Setup / PCR reactions per sample
Every
sample is applied to 3 (three) master mixes: MA
Bac for 16S, MA Yeasts for 18S,
and MA Control for the extraction
control.
All master mixes need at least one negative control and MA Bac and MA Yeasts have two PCR positive controls: P1 to verify the function
of the PCR and P2, a dilution of P1, as an indicator for the sensitivity
reached.
For MA Control there is no
positive control required.
Example
for the calculation of the required PCR reactions for the simultaneous analysis
of four samples below:
| MA Bac
(16S) | MA Yeasts
(18S) | MA Control |
Sample(s) | 4 | 4 | 4 |
Negative control (DNA-free water) | 1 | 1 | 1 |
Positive control P1 | 1 | 1 | 0 |
Positive control P2 | 1 | 1 | 0 |
Total number of reactions | 7 | 7 | 5 |
